There are two official ways to calculate a bonus at the end of the game.
To be eligible for this bonus, you must make the final move of the game by being the first person to play off all their tiles after the bag has been depleted of tiles.
In competitive scrabble tournaments, you take the value of your opponent's remaining tiles, double it, and add it to your own score. This is the method used by the National Scrabble Association according to rule III.C.1. of their official rules.
Or, you can take the value of your opponent's remaining tiles and add it to your own score once and subtract that same amount from their score once. This is the method used in the basic rule book that comes in the box.

Yes at the end of the game the total value of the letters in your opponent's hand are deducted from his score and added to yours and vice versa.
The other bonus is called a bingo which is 50 extra points for a word that uses all 7 tiles in the rack. This bonus may be collected anytime.

Yes, as Guru told you, if you end the game by using all of your last tiles. You get your score plus the total of the tiles of your opponents and they have to deduct the total of their remaining tiles from their score.

You can choose to either collected the points from the other players remain tiles, or they can subtract those from their score. It's up to the winner to decide. That's how I play anyways.
