All combi's have valves on the flow and return pipes just close these off and that's it done. release the pressure on the radiator circuit and do your repair.

The mains water shouldn't normally be running into the system, it should be sealed and only topped up from the mains if necessary. If however it is turned on there are usually two taps on either on of a (sometimes flexible) feed pipe. If these are off then it is isolated from the main water supply.

Underneath your boiler you will find a valve on the heating flow and return. These valvess are turned off by using a slotted screwdriver. Just turn them untilthe slot for the screwdriver is in a horizontal position. You don't have to tuern you main stop tap off to repair any heating pipework, although you will have to drain down the heating system to do any repairs.

It's a sealed, closed, pressurised loop, you cant shut it off. To release the pressure and stop the leak you will have to let water out somewhere, at the pressure relief valve or drain/cleanser tap. why not tighten the connection at the leak, that may be enough to stop it?
