If you want one soon, as opposed to months down the line when the Wii will be more plentiful, there isn't really "one place," it's a matter of timing. Get friendly with the people in the video game departments of your local stores; pay close attention to forums and gaming-oriented websites that circulate info and gossip on availability. It can be done. This holiday season, I snagged a Wii, a PS3, and a TMX Elmo, all at retail prices, all with no camping or hassle, just by paying attention to the grapevine.
The Wii, I hit upon on launch day by hanging out at the Amazon discussion forum that's part of its Wii product page. Several times during the day a new online store would make the Wii available, someone would post a notice on the forum and people would rush to that website (we wound up crashing quite a few), and some people would get lucky. Eventually I got lucky myself, when someone tipped us off to when Circuit City had their ordering page up.
The PS3 was thanks to a friend who manages a local Blockbuster--she tipped me off to another branch (a Blockbuster/GameRush) that had gotten five in. I strolled in the following morning and picked up the second-to-the-last one.
And the Elmo I got just by revisiting the Amazon Wii forums for old times' sake--someone posted about their sudden availability on kmart.com. Got one in time to give it to my little nieces for Christmas.
So getting these hot ticket items can be done without a lot of hassle--it just takes some good timing and good luck.

Wait till it is in stores again. eBay and Amazon.com have a lot of Wiis, but they are expensive. Way more than what they are really worth. I just contacted Nintendo about when they will be sending in more DS Lites and this is the reply I got:
"We're shipping Wii systems and Nintendo DS Lite systems to retailers
every day. However, we ship the system to the store's warehouses, not to
the stores directly. When they receive a shipment from they, they will
break it up into individual shipments to go to each store. They don't
share information about quantities or when those shipments will appear
in stores. For this reason, I can only recommend that you keep in
close contact with the stores for information about availability. "

Not yet. Currently Wiis are still in very tight supply and stores are getting just handfulls each week, if that.
It seems it will be at least a while before you can simply walk into a store and buy one for the listed $250 price. Perhaps 30-60 days away still.

You can get a Nintendo Wii at http://urlx.org/directtrack.com/aec4a and all you have to do is fill out the survey. I got my nephew a nintendo wii from that site and all i had to do was enter in my email address. If you really want one you should check out the site and get one from there.

there is no chance in my opinion to get a Wii soon in any retail store. As someone else mentioned somewhere all pre-orders will be fulfilled first. If any shop happens to get new stock imagine all the campers who'll sleep in front of the stores or the chaos and then within minutes all will be gone.
Another option - a very secure and reliable one which least know of actually - is to purchase a Nintendo Wii thru any trusted 3rd party seller directly at Amazon. They have good feedback, deliver on time, mostly are merchants themselves simply and Amazon operates as the platform, in fact the purchase is processed directly via Amazon as you would usually buy from them.
