I'd say Byzantium. England was quite a provincial island at the time, Spain did not exist as a united monarchy, the Holy Roman Empire was hardly a centralized state, and France was not as united and as strong as it would become later, after defeating English rights to French territory.

I'm gonna say the Holy Roman Empire would've been the wealthiest. France and England must have been pretty broke after all their fighting. I don't know anything about Spain. Not sure what you mean by the "largest" monarchy, but if you mean the most extensive network of royalty, that'd have to be the Holy Roman Empire, too.
