i am a girl .I have a friend who is a boy. We have been friends for last 2 yrs. He was very nice.
But one day, he touched me which no boy had done to me ever before. he holds my hand for silly things and often holds my waist when we are playing. Why is he doing this? How should i say that i am very uncomfortable with him?

He's not your friend. He is trying to get into your pants. Do not be delusional. He only wants one thing from you, and that is sex. He is not interested in your life or your charming personality or your opinions on current political events. He is interested in having sex with you. If you don't believe me, then ask him. You are in for a rude awakening.

I think it is normal to touch you if you have been in a relationship for almost 2 years. If you are uncomfortable with this behavior be sweet enough to tell him. 2 years, I think is a big period and both of you would have started understanding each other.
Come on try speaking to him.

Hi I think you need to be honest with him and tell him how he is making you feel. If he is a real Friend he will stop right away as the last thing a real friend would want to do is to hurt your feelings.

How old are you?? Don't you realize that he likes you as more than a friend!! DO NOT let him touch you if you do not feel comfortable! Talk to him and tell him how you feel...if he truly is a friend he will understand!

its not like that he is try to have s*x with u, but i guess he is likes u a lot and is making first moves that u get hints.
if u do like him and dnt like his touching,tell him frankly " i really like u, but i wuld apprecaite if u dnt touch me in the way u did all these days"
