I think one of the funniest scenes is the dead aunt on top of the station wagon in National Lampoon's Vacation.
The Baby Ruth scene in Caddyshack is also a classic.
Just any scene from There's Something About Mary is worth noting.
The scene in Me, Myself and Irene where Harry is trying to kill the sick cow in the road makes me cry with laughter every time.

Fart scene from Blazing Saddles makes the kid in me laugh every time. Zipper scene from "Something About Mary" makes me laugh and cringe. Diarrhea scene in "Dumb and Dumber" is great. Fake orgasm scene in "When Harry Met Sally" makes me laugh. I guess I'm a low-brow kind of guy.
