Yes, but not hard disk space.

In case that confuses you:

-Your computer has, most likely, many gigabytes of hard disk space. You install programs and have documents that take up that space. When a DVD is playing on your computer, you are not putting the DVD on your hard disk, so you do not take up space.

-You ARE using memory, though. Memory is magnetic space that is temporarily used by running programs and such. You have a certain number of, usually, megabytes of memory. Probably something like 128mb, 512mb, or 1gigabyte.
The computer stores upcoming frames of the DVD in memory and then quickly calls them to your screen and deletes them from memory. When you copy+paste in a document, for example, you quickly store the text in memory just temporarily.

The more programs you have open, the more memory you are using, so a DVD player is no exception. But it's not too much to worry about unless you don't have enough memory.
