Sandman, Venom (Eddie Brock), and Green Goblin (Harry Osborn). The "suit" takes over Peter, when he gets rid of it the suit consumes Eddie Brock and creates Venom. According to the trailers, Sandman killed Peter's uncle.

There are supposed to be three villians. First is the Sandman, second is Hobgoblin (Harry Osbourne), and the third will be Venom or at least when the symbiot seperates from Peter Parker. There was talk of Carnage in the next Spiderman movie.

Short Answer: Sandman, Harry Osbourne (called either Green Goblin 2 or Hobgoblin) and Venom.
Peter Parker does not become Venom. In the comics Venom is the entity formed between the Alien Symbyoite Suit and Reporter Eddie Brock after Peter rejects the Black Suit. Speculation is that in the movie the suit will be a new weapon/armor from Oscorp that tries to bond with Peter (In the comics the suit came from the Secret Wars Story and is later discovered to be a living symbiotic organism) When peter rejects the costume in the bell tower (the suit is sensitive to sonic waves) it bonds with Reporter Eddie Brock. Brock was a discraced reporter who was discredited by Spiderman and was goi ng to commit suicide - but first went to church to ask for forgivness. As the suit came off peter it landed on or near Brock and with their mutual hate for Peter Parker/Spiderman they bonded, creating Venom. This story seems to be intact to an extent in the movie trailer. Venom will appear in this movie.
The sandman was a smalltime criminal named Flint Marko who gained super powers by accident. The trailers make it apparent that Marko was tied to the murder of Peter's uncle Ben. This is a new plot device in the movie and has not appeared in comic continuity to my knowledge.
Harry Osborne is the son of Norman Osborne, the original Green Goblin. Harry does not know his dad was GG, but blames Spiderman for his death. In the closing minutes of the second movie, Harry discovers his father's secret room with all his Green Gobin weapons (armor, gliders, exlosives, etc) This seems to lead into Harry taking up the mantle of the GG or becoming Hobgoblin (another GG inspired villan from the comics)

This is pre-venom (introducing the suit), so Spiderman actually only faces Green Goblin (Harry Osborn) and Sandman.
Spiderman 4 will probably have Venom and possibly Carnage.

The answers above sum it up... Venom/Eddie, Sandman, and Harry/GGob/Hob (whichever he decides to call himself).
Read it a while back, but there was a rumor that Bruce Campbell's cameo in #3 is going to be the guy that becomes Mysterio... but if that's true, it'll probably just be an inside reference and not have anything to do with the main movie (like Doc Conners being in the 2nd movie but not being the Lizard).

I'm not sure about what the storyline is but I know that Spiderman is wearing a black suit in this movie. And from the end of Spiderman 2, I'm guessing he may be fighting against his ex-best friend, Harry, the Green Goblin's son. This should be interesting, can't wait!

I believe that Venom is played by Topher Grace (from The 70s show). This will be interesting to see him playing the villain.

Sandman, he is one of the new villains, and I think the suit itself is Venom; it is made of an extraterrestrial material that comes to life(at least that's how it went in the comics).

Spiderman 3 copied somehow the idea of Superman 4. The enemy is hero himself. Typical, but yet different stories.
