Take the Nagarcoil express...it goes via tirupathi and has a 2 hour stop there. You can hire a taxi and go and see Tirupathi and return in time before the train leaves or take another train back from that very station.

there is a direct train from mumbai via kadappa, ranigunta to tirupati. check google .com indian railway time table.
