A detailed instruction + example case would be helpful with the restriction enzymes involved. Thanks.

1) Find the restriction map.
2) Look for the various restriction enzymes available (e.g. Not I)
3) Check that your Gene of Interest and restriction enzyme sequence is different.
4) Insert your cDNA into the vector.

Example: Insulin gene into a bacteria factor using ECoRI as restriction enzyme. I did the same as I told you.
