You need to note on a few things. First, what your interest is in the company your applying to. Second, what you feel you can bring to the company. Third, the experience you feel you have that qualifies you for the job.
Basically, you need to summarize your resume into a paragraph. Be positive and show interests in the company your applying to.
When you have an interview, make sure you send a follow up letter to the person who interviewed you. Thank them for their time and tell them you hope to hear back from them.

For a cover letter - just go to your word processor and pick a style and fill in your and the employer's address, phone/fax.
Dear Mr. /Ms. /Human Resource Mgr
I am responding to your advertisement posted in the -----------------------, dated -----------------for the ------vacancy. Please accept my attached resume for consideration. Look forward to the opportunity of an interview with you.
Sincerely,
After sending, call the company to ensure receipt and restate that you are very interested, specify pertinent qualifications and then ask them when they will be scheduling interviews. About a week later call again. Interview = professional attire, respectful yet relaxed confidence. There is nothing you can't do or learn to do.

first thing, research the company. not intensly, just what it's about ie, is it industry, retail etc. also get the name of the manager or the person who deals with applications.(phone them and ask the name of the manager) . i would start the letter with, dear mr or miss etc, i would like to be considered for any vacancies you have for (whatever you want to do) ie, cashier, warehouseman etc) within your company. then say with x years experience (if you have relevant qualifications for the position then state them as well), i am confident i have the skills and ability needed to suceed within your company. enclosed is my cv for your attention. please feel free to contact me on (phone no) e-mail address etc anytime. i am available for interview at your request. i look forward to hearing from you. thankyou for your time. yours sincerely...............remember to paragraph your letter! you should get an interview if there's a job available. goodluck! p. s i know it's long but it needs to be. also write it out by hand in your own writing. DO NOT TYPE IT! it looks lazy and sets the wrong attitude.

Had pretty good luck in the past with letters to employers. So I would start with To whom it may concern: (Unless you know the persons name, owner, manager etc)
State your name & state how you are very interested in working for there company, (list preference of location if more than one) & state you wondered what position they may have open in any, & you would like to have an interview at their earliest convenience.
Then list how many years exsperience in one field, education, the position you want, than state however, you'd be open to any position they may have to offer (if you really want to work there.
name, phone number, email, & all ways of how to contact you, & that you have mutiple referances upon request
I'd give it 3 business days & call them to see if they received your letter & ask for an interview "Good Luck"

a LOI is usually written in Real Estate or contractors...but I suppose you could write one for a job. Usually just a resume and cover page summing up you resume and interest in the position is enough.
Type in google...resume and cover pages. You should be able to find something.
