I have a fender starcaster s-2 (remake of a crappy classic fender) It actually doesn't sound bad now that i have a decent amp. But its got 3 single coils and lacks the tone of a humbucker. Now since the space where the pickups are held is completely hollow, if i got a pickguard that would fit 2 of the single coils and 1 humbucker at the bridge would everything work out?

You can actually get humbuckers that will fit in the same holes. I put Seymour Duncan humbuckers in mine. I put humbuckers in the 1st and 3rd positions and put a Seymour Duncan single coil in the middle position. I also had an extra switch put in that switches the humbuckers to single coil if I want to get that single coil sound. Works great for me. Much better sound than the stock single coils.
