The fastest way would be a salt water rinse
Add salt to warm water and gargle for few secs, do this as often as possible, will help sooth your throat instantly!
2) honey and lemon tea
hope you get well soon! :)

I don't know about the best thing but I have used a solution of chopped up garlic soaked in water. Gargle it without the garlic. It is a natural infection fighter. Now what is the reason for the sore throat? That's what you really need to fix. Have you warn yourself out lately. Are you worried about something? That will do it. Get rid of the sore throat. Take herbs, homeopathic, or over the counter medicine for the cold. Lots of rest and lots of water. Then resolve the stress in your life. Personally I use mantra meditation for that. I get my best solutions to problems when I meditate.

Treatment
The treatment of pharyngitis will vary according to the cause.
Iodine compounds sold as Betadine or TCP can be gargled to shorten the duration of the infection.[citation needed] These compounds are effective anti-viral agents.
Antibiotics are only helpful when a bacterial infection is the cause of the sore throat. For viral sore throats, antibiotics have been shown to only affect the degree of pain by day 4 and shorten the average natural duration by 16 hours overall (14 people need be treated for a week to reduce the duration by one day).[1]
According to Traditional Chinese medicine, pricking the acupuncture points Shangyang L.I.-1 and Shaoshang Lu-11 and squeezing out a few drops of blood resolves most cases of acute pharyngitis, tonsillitis or strep throat within 1-3 days. Most patients feel significant pain relief within minutes of this procedure. [5]

This is what I do (I try it all):
1) Cut up approx. a teaspoonful of fresh ginger, and chew on in. Drink the juice from it but you don't have to eat it. That gives your body strenght for healing and it burns good.
2) Gargles with Listerine to kill bacteria as much as possible.
3) Gargles with Warm water and salt to kill the other bacteria.
4) Hot herbal tea with lime and with honey. Honey is good too.
5) Hot shower or bath with aromatic soap
6) Tons of Vicks Vaporub on chest and neck.
7) Most of all, restful sleep. Try even some Nyquil if you are Ok zonking out for the night.

As awful as it sounds, mix apple cider vinegar and honey, and then drink it. Yes, drink. Not gargle.
I was wildly doubtful when my SO recommended it, thinking it sounded way too freaking granola, but it really is almost a magical cure-all. The web also recommends it: do a search for "apple cider vinegar" and honey. Listed a few results in sources.

hot tea with lemon and honey. Honey soothes the throat and the lemon has vitamins in it as well as some antiseptic properties.
