Anne Frank generated a moving story by simply writing in her diary. While the story was never planned, it had all the elements of a story. Some are saying the theme is "the Holocaust" or "WWII" but that is the setting -- not the theme. We know the ending which ends in irony and in that we can assign meaning where Anne Frank never intendended. We can see how Anne Frank went through a change throughout her writing. Essentially she became a prisoner, alonger with her family, in the home. But she finds happiness in living day-to-day with her family. So although she was a prisoner, she was truly free in this routine -- she still had her family to shelter her. So, the change that she goes through in this imprisonment is essential to determine the theme. I would say the value that changes is freedom and state the theme as: We are truly free when we live day to day. I suspect when her family was taken away everything changed and that confirms the theme.
While all stories have multiple themes, the question presumes we're searching for the main theme. The main theme is the one that is found throughout the story, not in sections.

Your English teacher asked this and should be bludgeoned for it. English teachers do everything they can to screw up enjoyment of literature.
Anne Frank was a young girl, not a professional writer. She didn't set out to have a theme. She recorded her thoughts, experiences, feelings, and observations in a private diary. Would that the Nazis come and take away your English teacher.
