This varies but for me...
....someone who has no or low self-esteem, who continually bangs themself up over everything, no confidence, no desire, no inspiration, no passion for life and ultimately no yearning to change any aspect of their dismal outlook.
On some level they must enjoy the misery as this is what they have chosen. Even if external circumstances have dictated alot, how the individual chooses to act/react/behave says so much.
All up....Someone who believes they are a loser, will be a loser because this description is embedded in their psyche.

I think it is relative. Someone who does not meet your expectation is a loser. It can be financial, social grace, lazyness, etc.
What do girls mean when they say a guy is a loser? Well I'm no girl but I'm thinking they mean that he is just not good as a potential mate. I guess this would be the Darwinistic version of "loser" :)
