Anti-spyware is much easier but if you want to do it manually boot in safe mode or better still from a live CD then edit the registry to remove all references to the spyware files then delete the files, you may need to replace infected files with clean versions, then check the other windows startup files for references to the spyware.
If you are not sure what to remove you should have used anti-spyware.

To remove spyware with a program, you would have to know everysingle file the spyware uses/has and manually remove them including any register entries as well.
The better and smarter choise is to use spybot or adaware to remove spyware.
If you're talking about a virus, only way is to format and reinstall or get an antivirus program. There are a few websites that will scan and remove viruses.

This would be very hard to do without aid of an application. Behind each antispyware program is a team who researches and discovers new forms of spyware and how they can get rid them. This is why there are updates to the definitions file which contains signatures of each spyware form so it may counter them.

Not a good idea since you don't know where all of them are in your system. Even some anti-spyware apps will have problem with the spyware. There are quite a few anti-spyware apps that you can use and they're quite effective. They're FREE too. You can also install more than one anti-spyware apps on your pc. Here's my recommendation:

1. There are free programs like AVG or Avast who should clean ur computer. Better ones are paid for. BitDefender and Kaspersky are best in this field. 2. Also check if you have system restore point before u got this spyware. You can go back to this restore point. This may help.

You can't because you can't find it all without reformatting your hard drive, and even then, you will drag the virus right back in with some of your files, since you don't know where it is. Get antivirus like the rest of us, run it and keep it up to date and you won't have this problem.

It is doable. But let anti spyware doit, it's (for lack of a better term) mechanized.

Very difficult. Sometimes you can delete the corresponding file manualy but sometimes it is inside the OS. You need an antispyware to delete it for you. I recommend the two best :
-Kasperky (pay)
-Ad-Aware (free)
