Following a male's engagement of a female and given even only 5 or 10 minutes to recover, if this girl really sets him alight, he can re-engage her physically, and if he is excited sufficiently by her, he will produce even more sperm even while he is doing her a second time.
And if he really 'just loves it' with her, his explosion will be just as great, even if the sperm count is less.
But, the point is, if he really had a good time with her, and left the scene, he would be thinking about her over the next 2 or 3 hours sufficiently to get a solid hard-on quite a bit, and, therefore, reproduce sperm quite quickly in the old 'ball factory', if he is very sexually motivated by her.
So, often, even only 6 hours later, it would be very hard to tell if he had recently had sex, even if you did a sperm count on him - especially if he had thoroughly showered, as you would probably not even detect DNA presence either.
I think dgriffith had the best possiblity.
If he has had a very good time with her - not just sex - he will have a glow about him that will last for some hours.

I liked dgriffith's answer. However, no, there are no actual scientific ways to test if a male has had previous sexual intercourse unless he contracted certain sexually transmitted diseases though intercourse.

This question is hard to answer, these days even female, you cannot tell, as they currently have hymen repair surgery. As male, we also had wet dreams, not intercourse, but similar. Thus, the answer would be no.

Lie detector is the best choice. But if you have suspicions on who he might have been with, you could ask her.

Well, you can test him for female DNA to see if he had a recent experience... look for any rests of 'foreign' bodily fluids on his body.
Otherwise, no.

no way to know if old one, but after intercourse with one day, check his balls empty or full with semen
