I go to Las Vegas 2 - 3 times a year. I was there two weeks ago in fact, at the Luxury Travel Expo.
Since you asked what my favorite hotel is, not the best value, cheapest, etc. - its an easy answer - The Venetian wins, hands down!
The rooms, the restrauants, the entertainment, the ambiance, the shopping, the attractions, the entire feel of the resort is unlike any other.
We stayed in Mandalay Bay on our last visit - great resort - terrible location! It's the last hotel on the strip, and even with the free monorail to the Excaliber, it's just too far away from where we like to be. The rooms here are just OK, but they are in the process of a massive room make-over. Still, they don't compare to the (standard!!) 750 sq. ft. rooms at the Venetian.
The visit before we were at Bellagio - great hotel, great location, great big bill - but we enjoyed it. The best rooms here face the lake, so you can watch the evening's performances of the fountains - you get the music through your TV!
Here's a little tip for you - stay at the big hotels but gamble at the little ones. Our favorite gambling spot is Casino Royale - a little hole in the wall stuck between the Venetian and Harrah's. The dealers are much more friendly there than in any of the large hotels, and my wife says she does better at the slots.

On Strip try Bellagio or Mirage. Off Strip the Palms(pricey) or Rio Way off Strip Red Rock,Santa Fe or one of the many Stations- Oops, the Red Rock is a Station Casino! Downtown the Golden Nugget or LasVegas Club. (the golden gate is city's oldest>100 years old and worth a look.) You didn't mention price as a consideration so some or most can get steep, especially over New Years. P.S. don't miss having a drink or partying the night at the Doubledown Saloon. Caesars Palace was awesome!!! My sister got married there so we all stayed. It was awesome. We had the cheap room there and we still had a hot tub in our room facing the strip. It is a huge hotel so there is lots of shopping, dining, and entertainment. There is a mall right in the hotel! It is pretty centrally located on the strip also so you are able to access other parts of the strip fairly easily. The pool area is also gorgeous - Have fun - When it comes down to it, you seem to spend more time outside of the room anyways so it may be worth it to stay at a less expensive hotel.

Mandalay Bay, I stayed there last summer and it was great. The place is huge and connects directly to the monorail to NYNY so some walking can be cut out. The pool area is immense with an actual beach-like setting with waves, lazy river, and even a nude tanning area. Plenty of places to eat and do the sin city thing. The bathrooms are huge there, often with a jacuzzi tub and separate shower stall. I can say the bathroom was just as big as the actual room.

The Mirage. It has it's own unique scent throughout the entire hotel that gives the feeling you're in a tropical paradise in the desert. Their pool is awesome and the lobby is gorgeous.

My favorite is Aladdin. Great location. Love the attached mall. The spa is fabulous. And the buffet is real tasty.
I agree with Travel Guy, too, Casino Royale is great. My husband loves to play craps there for the exact same reasons.

Just came back - stayed across the street at the Mirage. Spent most of my time at the Venetian.

I stayed interior the inn for one evening and that is stunning. each and each room interior the Aria inn is better with unique paintings and a range of books. there is also a flat reveal computing gadget in each and each room and is equipped with the biography of the featured composer or artist and an iPod connection loaded music of fifty 2 composers.

Depends on the price.
High end: I like Bellagio, Venetian, Wynn's
Cheap end: Monte Carlos (short walk from my car to lobby)
