No, Donald Duck was created by the Walt Disney Company, Daffy was created by Warner Brothers.

Not any more than any two human beings being related. However if we take "Who Framed Roger Rabbit?" as cannon, who knows.
Another question might be is Howard the Duck and Donald Duck are related. They both have the same last name, similar coloration and seem to both come from worlds where the duck is the dominant species, as opposed to Daffy who comes from a world of mixed species (including hairless apes better known as humans),
