A serial killer is someone who commits three or more murders over an extended period of time with cooling-off periods in between. In between their crimes, they appear to be quite normal, a state which Hervey Cleckley and Robert Hare call the "mask of sanity." There is often — but not always — a sexual element to the murders. All the murders must be completed/attempted in a similar fashion or the victims must have something in common, ex. occupation, race, sex, etc.

A mass murderer, on the other hand, is an individual who commits multiple murders in a single event and in one location. The perpetrators sometimes commit suicide, therefore knowledge of their state of mind and triggers their actions is often left to speculation.
In 1927, school board member, Andrew Kehoe, upset by a property tax that had been levied to fund the construction of the school building, killed 45 people (including himself) and injured an additional 58 in a bombing at the Bath Consolidated School. This is still the largest school massacre in United States history.

i spsoe we will never know the answer to that one, there is a marine sniper during vietnam taht had over 100 confirmed kills papa something, there were also japanese officers during ww2 and sepcifically the rape of nanking that had contests on how many they had killed i beleive they were in the 120s. john wayne gacy claims to have killed over 150 tho many less have been confirmed, timothy mc veigh killed 168 w the oklahoma city bombings, mohammed atta can be said to haev killed 1/2 of the people in the WTC disaster or roughly 1500, not sure who the other pilot was

I am not sure if their was ever a historic record of how many people died in wars hundreds of years ago but I found this quote
"These be the names of the mighty men whom David had: The Tachmonite that sat in the seat, chief among the captains; the same was Adino the Eznite: he lift up his spear against eight hundred, whom he slew at one time..."
If he did that in one day, he may be the person.

I'm not sure. I would guess it would have to be a serial killer, which is a person who kills many people over a period of time.
John Wayne Gacy officially killed 33. I'm sure someone out there has out done that.

Considering the number of unsolved murders, the cleverness of some serial killers, it is probably a person unidentified.
There are lots of successful murders, and even with computers, crimes by one individual are often not associated until they are caught, if they ever are.

Fortitude... Could have been the bombadier on the Bock's Car -- Nagasaki. Of course, if you want to look at team effort. I'd have to go with the night the U.S.A.A.C/U.S.A.F. dropped incendiaries on Tokyo or Dresden.
