As was said previously, you need to play multiplayer for a combined 20 hours of human time.l The best way to do this is to set up a four player battle (no cpu) with one stock, infinite time limit and no items. Then just leave it on overnight. This will be enough to put the combined human hours to more than 20 hours and you will then get the chance to challenge Mewtwo once the match is over. DO NOT RESET THE MATCH OTHERWISE THE HOURS WILL BE RESET

Play 200 hours of Multi Player, with 1 human in Melee mode. A good way to achieve this is to leave your Nintendo Gamecube on for 24 hours (or over night if you play the game a lot in melee mode). After the 200 hours you will have yo battle Mew Two and when you beat him you will have him.

To unlock mewtwo you need to rack up 20 hours in combined Vs. Play or play 700 multiplayer matches to face him. Beat him and he's all yours.

turn on multi-player mode and leave it sitting there for 20 hours it says when you stop that mew two is unlocked
