Does any desktop hard drive work in any desktop? I need to find out where I can get a second hard drive for my Emachines T6216 (I need two of them. :)

No, not any desktop harddrive. Hard disk drives are normally connected to SATA, SCSI or IDE controllers. You need to use one that is an IDE drive.

Any will work provided it's the same type as the connectors inside your PC.............. usually IDE, but increasingly SATA / SATA II is becoming more popular

Or you can also pick 'em up on Ebay... make sure it's either a SEAGATE, WESTERN DIGITAL or a MAXTOR as they're generally the best

No there is SCSI, IDE and Serial ATA you need to find out what you have.

that machine takes IDE HD so just make sure you get a HD that is ide capable.
