The only way I know of is to totally empty the area under the sink. Put a bucket under the pipes and go to work. Other than that. If you have a compressor wrap a rag around the air chuck and stuff it down the drain then blow the water out of the pea trap before you start.
Good Luck. Make sure you have some towells ready. LOL

i understood your ? differently than the others. under the sink, below the bowl on the back is the lever for the stopper. remove the nut around the shaft and slide the shaft out. that will allow you to remove the stopper from the shaft. when you put it back together, don't engage the stopper to the shaft. i didn't on mine and it still works fine, plus i can take it out when i need to. hope this is what you meant. good luck, and happy holidays!

Are you trying to permanently remove the "pop-up" assembly? If this is your question, then go to a plumbing supply store, and ask for a "grid strainer". It is a one piece assembly, that fits in the same location as the pop-up, only it has no moving parts. They are used mostly in commercial buildings, so that vandals don't stop up the drains in public restrooms. They cost about $15 dollars, and are chrome plated brass. They are very durable, and only minuscule items lass than a 1/8" can fit through the holes. I hope this answers your question.
