No way can I pick a funniest movie, but I will say I have never seen a Mel Brooks movie that disappointed me. As for top one-liners, no one has mentioned the line spoken by a middle-aged woman after Meg Ryan's faked orgasm in "When Harry Met Sally": "I'll have what she's having."
By the way, I think that line was delivered by Rob Reiner's mother, but I won't swear to it. Considered how often this line has been plagiarized (for example, in a shampoo commercial where a woman is screaming "yes! yes!" after shampooing her hair), this line definitely deserves a place in the One-liners Hall of Fame.

The funniest movie ever made is Richard Pryor: Live in Concert (1979).
The funniest one-liner I can remember at the moment is Hugh Grant's first line -- actually his first twenty or so lines -- in Four Weddings and a Funeral. It begins with F and ends with K.
I thought There's Something About Mary was pretty damned funny too.

Yeah the Ringer is such a funny movie although it is a bit harsh on the special people. Borat is a funny one if you've seen it.
Funny quotes from both of these movies -
The Ringer ; "Guess how many fingers I'm holding up?", the other guy says "10" and then he ^ says "No, 8 - 2 thumbs and 8 fingers" and they both laugh their heads off! LOL
Borat ; " Hi, I like your wife! High five!!"

Don't Be a Menace to South Central While Drinking Your Juice in the Hood
The title is pretty long but the movie (produced by the Wayne's Bros.) is funny b/c it touches on the stereotypes of black community. Def. a classical.
Also Any Will Ferrel Movie (ie ELF n Kicking & Screaming) :-D

you will no longer be able to take it too heavily. it quite is what it quite is - a slacker's moist dream. It replaced into between the dumbest movies of the year, yet possibly once you're interior the the suitable option temper you will come across the fairly some throwaway strains humorous.

There were no words involved, but the act itself said a thousand words.
It was the snowball scene in "Dumb and Dumber".
( You must see it!! You will laugh so hard.)
As for one liners I would have to say in Happy Gilmore.
Sandler comes home and finds his girlfriend cheating on him with another guy. The guys comes out of the bathroom. He's 70!!
He looks at her and says. You're cheating on me with this guy?
She replied. Yes, he's got a 5 year plan. Sandler replies, what? live.

I was beaten to it... Blazing Saddles is the funniest.
UPDATE: I changed my mind -- The Big Lebowski.
The funniest line from that movie:
Walter: You want a toe? I can get you a toe, believe me. There are ways, Dude. You don't wanna know about it, believe me.
The Dude: Yeah, but Walter...
Walter Sobchak: Hell, I can get you a toe by 3 o'clock this afternoon... with nail polish. These f------ amateurs...
My opinion of the best one liner of all time is "Hi. I'm Bob. Would you knock me out, please? Just hit me in the face."
That's "What About Bob", obviously.
