When you pierce something you are essentially puncturing your skin. If you leave something in the puncture (an earing) it will prevent your skin from healing (growing back together). If you remove an earing from a newly pierced ear, the skin will heal (grow back together). The amount of time this takes will vary from person to person, but normally, it won't take long.

If it's newly pierced you shouldn't take it out to clean it. Closing can be anywhere from a few seconds to an hour or two, depending on how long it's been pierced and where. If it's your catilage...expect a few seconds to a few minutes.
If you need to clean a new piercing, try rotating it either with the stuff they gave you when it was pierced, peroxide or rubbing alcohol. Those last two shouldn't be used if you have sensitive skin, with some people they make the piercing worse.
If you try to put an earring back into a closed up or closing piercing this will irritate your ear and make your more prone to an infection from irritating it. So if it does start to close, soak your ear in some warm water and epsom salt for a few minutes, this will help clean and soften it, then if you have a piercing stud, it's sharp like a needle...put this through the hole and leave it alone until it's fully healed. 6-8 weeks for lobes. 4-6 months for cartilage.

In 2 hours they might not be completely closed, but I wouldn't risk it if you can help it - being its gunna hurt to re-punch it through or even to get it back in with the raw tissue. If it's a necisity to take it out for awhile because to the metal (ie. hospital stay or tests) you can take a needle and attach some string to it - then insert it into the ear - but not with the pointy end!! then tie the string, that will keep the hole open for a few hours, though I wouldn't recommend it for extended periods.

They close up very quickly... within a couple hours. My friend has put fishing wire through the piercing holes to keep them from closing. I haven't tried it, but she has several ear piercings... not just the earlobes... and it has worked for her on many occasions. Good luck!

it will began to close with in a few hours. I had new piercings when I went into the hospital for an appendectomy, I had to remove them before surgury and by the time I woke up in recovery (5-6 hrs later) I could not get them in.
