It depends on the definition of city.
In the United States there are legal definitions of the term city. For example, in the state of Indiana where I grew up a municipality can elect to become a city if it has at least 5,000 people. But there are many towns that are larger than this that remain a town. The difference is the style of government, usually that a city has a mayor and a council, whereas a town has a council that hires a town manager.
I live in Illinois now in the "Village of Glen Ellyn". Villages have a legal definition in Illinois and Ohio. Indiana only recognizes towns and cities.
If you are calling a city a place on the map, there are probably many "cities" with a population of zero, ghost towns that have gone away for a myriad of reasons.
Looking on the web there is one surprisingly small but very famous city: Vatican City. Prior to 1868 Italy had many city-states until becoming a unified country under Vittorio Emanuelle, the first president.
Two sites below, finetravel.com and Wikipedia list its population between 600 and 700 people. On Wikipedia the author goes as far as listing the jobs of its 557 inhabitants as of 31dec2005.

Vatican City, located in Rome, Italy. It has the smallest population in the world, but the curious fact is that it also has independence as a country, for it has even its own banner, so it acts as a city and a country, or independent state, the population is aprox 1000 people.

Intersting question, i wonder why u asked sick of humans? Want to get away but have hospital close? There are a few places left u would look for a hard to find area rainforest ,a waterfall in a cave, Tennessee has a lot of places with few people of corse there are snakes. a deserted island maybe.If they are not all bought up by now. also how would u eat? thats really a hard one punch up least population in us on the internet

The city of Carthage (the one in Africa, not the one in Missouri) has zero population. The city of Pompei has zero population. The city of Troy (the one in Turkey, not the one in New York) has zero population. Uxmal has zero population.

One of the requirements to be classified a city is that the population must exceed 25,000 people.

Well there is this place called the MacDonald islands that is a nature preserve that only a few scientists live as well as some birds and seals but i don't know if that would count as a "city"
