I'm going to go against most user responses here and say Speedy Gonzalez because he's smarter.
Here's a dialogue with me and the Road Runner:
N: Ok, I want to start the race now and need you to get to the starting line.
RRunner: Beep... BeepBeep
N: Look, I'm trying to get this race going, I need you to get on your mark. Tons of people are watching...
RRunner: BeepBeep, Beep...
(Speedy Gonzalez, sitting in the shade, just looks at me and shrugs... I move closer to The Road Runner to position him and as I do so, he runs off. For some reason a Large Boulder falls on me...
Great... now I'm Wile E Coyote!! I hate when that happens!
Speedy laughs!
I'm finally able to get Road Runner on his mark and the sound of the pistol gets him going but he veers off and heads in another direction)

ps (edit): In the real world, the length of the legs have little bearing. This is true for sprinters, marathoners, soccer players, etc. In fact, many times, it's the other way around.

Technically, the road runner would be faster since he has longer legs.

as with all things such as this, when they have raced it ended in a tie or draw...
but looking at the cartoons, road runner appears much faster than speedy...
