Depends on the type of bird. My parents had starlings build nests in their roof and patched them up but they just pick through the patch. In this case you might have to get animal control or if you are out of town shoot them.

Make sure you get ALL of them out including the nest and block up the the access holes they are useing. Wire mesh, chicken wire works good. Cut it to size and nail it up and they cant get back in and will have to move to a new place to live.

Try to watch and learn when they go out to feed.
Then close up the hole with mesh.

Good Luck on getting them all out. We had squirrels in our attic. Lets just say, dead squirrels in the attic are a very bad thing.
