After one ejaculation it takes about 3 days for the glands to return you to a full load. However, just a few hours later and another ejaculation and you have enough sperm to fertilize a lady.

For a full load per se, 2 to 3 days is enough. After 3 or so days, the excess semen is evacuhated from the body through excrement and urine.
Semen is a fully renewable resource. When you ejaculate semen, your body simply makes more — the same way your body makes more saliva after you spit.
