I suppose the easiest way is physical examination to see if 'she' has a vagina!!!! That said, the "hole" should be a true female organ as transverstite can have a hole literally punched into that area.
Further investigations can then follow by testing the testesterone level

it depends on whos playing who..... i am a sporty chick and i love doing guy stiff buti am the most girlest chick i know. i cant stand when dudes say well your a girl you cant beat me...well think again. girls are just as strong as guys. have a merry christmas.
