Pests are killed with pesticides, and pesticides are poisons! So, our yes to your question is a qualified one. The lay person has a tendency to spray until everything in sight stops moving. In so doing, two problems arise: 1) we tend to overkill the insect population, and 2) the unnecessary application of too much chemical can be dangerous to children, pets and plant life. Chemical spraying is only a small part of the work required to do the job properly.
A friend of ours at a garden center tells us that Ortho Chemical Company strongly recommends the following procedure: remove loose wood, trash and clutter from areas where spiders might hide. And, wear gloves and protective clothing when cleaning up infested areas. Also, vacuum infested areas to remove egg sacks, and destroy the contents of the vacuum cleaner bag.
Spray outdoor living areas and cleanup debris around the home that may harbor spiders. Kill spiders by spraying webs and infested areas with Diazinon Insect Spray, or dust areas where spiders might hide with Ortho-Klor. The active ingredient in Ortho-Klor is Dursban, which has been marketed as a replacement for Chlordane.
By the way experts tell us that pesticides should not be discarded, but kept until they are completely used up. The philosophy here is that if they are dispensed a little at a time they tend not to be thrown back into the environment in concentrated form.-

I'm not sure about the least effective way to kill a spider, but I can share what my grandfather has always suggested as the most effective way to kill any bug. You convince it to lie on its back. then you start tickling its belly. When it starts laughing out loud, you put some DDT (extremely deadly insecticide) in its wide-open mouth.
Done.

First of all i am not sure if your question should be LEAST effective or MOST effective way of killing a spider!
LEAST- feed it with insects and keep the whole place cool.
MOST - whack it soundly and it usually dies.

(-: Scream at it and run away. Offer it some curds and whey. Wash it down the waterspout. Invite it into your parlor.
(-: I try not to be superstitious, but spiders kill bad bugs and I like to hedge my bets, so I try not to kill them. Instead, I grab them in a tissue and throw them out the window. Maybe they'll live, maybe not.

you want the least effective way? if you don't want to kill him, just splat the floor beside him, that won't kill him, or just spray with buy killer around him, and he will surley walk away.. He will never forget that you spared his life LOL!!
Leave him alone and he'll go home !!

I always put a glass or something like that over it and wait for it to either suffocate or die of age. It sometimes takes a long time, but it's better than having to feel it squish.

I use Morteen on it .. i keep spraying ... and spraying ... and Spraying until it is all covered in what looks like snow .. but it still moves
EWWWWWWW I hate spiders ..
Just STOMP on it!!
