You would take the MCAT during college, most likely your junior year before applying to medical schools your senior year. If you were to take a year off after undergraduate before going to medical school, you could take/re-take the MCAT during your senior year.
Because the MCAT costs a decent amount of money and a lot of studying, most people take it after finishing their pre-med course work (organic chem, general chem, biology, biochem). However, it isn't required that you finish these classes before taking the test.

during or after college, near the end of 4th yr or 5th yr. there are lots of require classes to take before you can sign up to take MCAT
