Take my advice and take Heathrow express to London. It is a train, but not any train leaving from Heathrow, ask for Heathrow express. It takes 20 minutes. Traffic is often quite bad and it could take you up to an hour by car. Let alone the cost. Heathrow express cost 15 pounds I think, I can't even guess what a taxi would cost!
Heathrow Express ends at paddington. From there, take the tube (underground) and you can go :
1. To Oxford street and Bond street for shopping
2. To Notting Hill for a nice walk, and then to Westminster Abbey and St. James's park, very nice and calm
3. To Covent Garden, Piccadily circus and Leicester square, to get a good idea of London atmosphere
4. Combination of the above, if you are really fast!

Dont even THINK about getting a taxi if you only have four hours..it will take at least 40 minutes to drive into central London, and very probably longer depending on the time of day.
Fastest route is the Heathrow Express train to Paddington station, takes about 15-20 minutes non stop.
But, to be honest, you really aren't going to be able to get much seen or done in only 4 hours...

If you mean you have 4 hours between landing and your next flight taking off, then forget going to London, you will not have time.
Bear in mind that if you leave the airport you will have to go through immigration which can take ages and you will have to be back at the airport in time to go through security and get to your check in gate, which again can take ages.
To answer you question tho',allow 40minutes by tax. Youuse 'down town' which is not understood in the UK but Ibelive refers to the financial district (?) Do you really want to go to the financial district?
London is huge -- make sure you know exactly where you want to go and see. The official airport website says allow 60 minutes by taxi and £46-£50. 4 hours is going to be hard. But don't take a taxi because it will cost you a small fortune. Take the Hethrow Express, around 14 pounds and you will be to Paddington Station in 15 min.
