Not in general but I beleive that if a guy is sexually experienced and it is a girl's first time, then maybe the guy could tell by how she responds to the sexual activity. That's just an assumption.

if a guy is experienced in deflowering a virgin than yes. if he's never had one than probably not. inside you is thin membrane of skin that rips open when you have intercourse. most women bleed their first time i myself didn't. so that's how you know

Your average guy won't have a clue unless you tell him.
