I think you are confusing the two terms. You create a partition of space on a hard disk and than format that space so that the Operating System you are using is compatible with it. For example, if you are using Windows XP you can format a partition for FAT16, FAT32, or NTFS. If you are using Linux you would still create a partition and format that partition under a Linux addressable filesystem.

There are programs out there such as Partition Magic which can resize a partition without erasing the data, and you would not have to reformat the partition.

1) Jaycetan doesn't know what he's talking about.

2) Your harddrive MIGHT be partitioned for a reason. You may want to find out if you really SHOULD de-split the partition before doing so

That being said, there are PLENTY of

"Partition manger" programs [try googling that term]

Symantec (norton)
Powerquest both make popular products.

here's the cavaet.

if there is even the SLIGHTEST bit of problem (or if you don't know what you are doing) the chance of DATA LOSS is quite severe.

I would NOT "unpartition" without first backing up your system

Why do you want to unpartition it? the best way to save data is to put data on a second partition that way if you lose the MBS (master boot sector) you don't lose your data files, Just reformat the first partition reinstall windows and hey presto you have your data files... plus the manufacture may have installed the system back up files there and if you lose that you will have no way of reinstalling your system
Your choice

Yes, you can do that with 3rd party software like Partition Magic or Acronis Disk Director. I prefer Disk director because it is easy in use and has a friendly interface. Disk Director can resize, move, copy, split, and merge partitions without losing your data. Follow the link for more information:
