Both are acceptable. You will find that "friendliest" is usually used in a descriptive form, such as: "They are just the friendliest people!".
Whereas "most friendly" is usually comparative,e.g."They were all friendly, but John was the most friendly".

Depending on the context, "most friendly" would be acceptable, but is a bit more awkward sounding and more of a mouthful.
