Like the previous poster said --

You set up a proxy server for your users, which has the ability to filter web pages. Squid is one such example.

There is a file in your Windows directory:
c:\windows\ system32\ drivers\ etc\ hosts
(Windows may also be in c:\winnt or some other directory)

At the end of the file, add:
127.0.0.1 domainname.com
where domainname.com is the domain you want blocked. You can add more of the same line for each domain you will block.

The entire domain will not appear on your computer. I don't think this will work for individual pages, but ads are usually served from specific servers that you can now block intentionally.
