Bond was joking with her. The word was "Mom," which he did say as he was leaving in the elevator.

In the books, M was a man, of course. His name was Miles Messervey.

Q, by the way, is short for "Q Branch," the armorer division of the secret service in the films. Q's title and name are Major Boothroyd.

Bond creator Ian Fleming chose Boothroyd as a nod to a gun expert who wrote to him after the first few Bond novels to say that Bond wouldn't use a Beretta in the secret service but a Walther PPK.
