It depends entirely on where you live. The Tony's are indeed THE award for American theatre, however, here in Canada, the biggest award is called the Dora Mavor Moore Awards (aka The Doras).
There are also several hundred different types of awards and honours given to productions across Canada. Again, it depends on where you live, what association you are with and the genre in which you work.

The Tony's are the biggest, but I can't believe no one's mentioned the Drama Desk Awards! Those are also a pretty big deal. Then there's the Pulitzer category for plays, but that is based on a script, not on the actual performance.

In Chicago, there are the Jeff awards, which are given out to mostly storefront theaters. I'm not sure of the rules to qualify to be judged by the Jeff committee. There's also the After Dark awards that are given out by a local gay magazine.
