Depends on the model of car, on most the engine is running with the car in park or neutral it usually is written on the dip stick

Turn the car on, go thru all the gears, (Park, Reverse, Neutral, L1, etc.) one by one, go back to park leave the car running and check the level.

You should do it with the engine on, for better accuracy.
Some dip sticks say COLD or HOT, that means, engine OFF or ON.

on, further more you should drive a few minuet before you check it as well, or with the car stopped and your foot on the brake switch in to each gear stopping for a second or two to allow trans fluid to go into all parts of your trans mission

on the dipstick it will read " check while in park and engine running" Do this on a level surface and on the dipstick it will also tell you what transmission fluid to use.
