Brother and sister have the closest DNA match because they're coming from the same parents (which similar in DNA).

note: sorry, now that i rethink again, i think mother and child has closest DNA match....Sorry for the confusion.

While a child gets half of its genomic DNA from its mother and half from its father, there is also DNA in the mitochondria which are only inherited from the mother.

Generally brothers and sisters are considered to share about 1/4 of their DNA.

Hopefully husband--wife have minimal match (i.e. they're not first cousins)!

mother child is greatest, i guess.
but, father child is also has close match but less than the above choice.
it's because
1. the fertilization takes place inside the mother's body.
2. we are breast-fed by mother

3. our mitochondrial DNA is inherited only from mother.
hence, mother-child is a closeset match.
