Please tell me what is 6X His tag. And where is it used? Biologists please come and answer this.

If you want to purify easily proteins you put a tag either at the N- or the C-terminus. The tag can be from a short sequence to a small protein. The way you add it is by cloning the respective DNA sequence fused with the gene of interest.

The 6x His tag is a short tag consisting of 6 histidines in a row.
Since it is a small tag it has the advantage that it usually doesn't interfere with the protein function at least like bigger tags like GST would.
It forms a complex with Ni2+, so it can be used for purification of the desired tagged protein with affinity chromatography with Nickel columns.

There are also antibodies against the tag, so you can use whatever immunochemical assay (e.g. Western blotting, protein localization, etc) you want to, without needing to raise antibodies against the specific protein.
