NYC is the best and then Chicago. What they have in common[ unlike many American cities including LA ] Is that people actually live in the city-all don't flee it after working hours.Middle class to very wealthy people live in these cities which leads to a flow of commerce and culture .
Money stays in the city and the city remains healthy and vibrant because of it.
In addition all great cities are walking cities and NYC is the best because it has so many great places you can walk to.
To that you can add the fact that NYC is not only the most culturally diverse city in the world but also the most tolerant city of it's size and the result is the best American city.

If you are refering to an area of more than 100,000 people. Then the any one you go to is the best. All American " cities" have many things to do and see. The differences are those of geography, weather, and the history of that area. Best is here having a memorable experience. Worst is where you try and forget your experience. That means your experience determines the " Best".

my home, north branch, mn. but actually ny is the best. you have such a concentrated mix of culture and yet they harmonize (most of the time). did you ny central park is man made? all the dirt and vegitation was hauled in and the footpaths were designed and yet seemingly natural.
