I ran a letter through a Pitney bowes machine at work and dropped it in a box at the Post office that had a small sticker that said "stamps only" will the letter get to its destination.

Yes, USPS used to separate metered and stamped because the machinery they used to scan & cancel were different. They now have a single machine that does both. Even though the drop boxes may still have the stickers on them you can put mixed letters in the box.

Good question. I'm guessing stamped mail. In reality people put the wrong stuff in those mailboxes all the time so it gets sorted anyway. I put the no postage required in the stamped mail slot though unless there actually is a metered stamp on it.
