What would define best? Looks? If its looks the antique Bugatti's and Mercedes are very beautiful cars, the Bentleys and Rolls Royce's have very graceful smooth lines. Power? If you are looking for bonecrushing power/speed Dodge Viper, Chevrolet Corvette, the high end Ford Mustang's like the Saleen and Roush are tough to beat as well as the Ferrari and Bugatti Veyron.
Luxury? There are plenty of cars that give a great balance of power and luxury BMW, Audi, Infinity, Lexus and Mercedes all offer models that give a fantastic balance of power and refined luxury.
Practical? There are many cars that are all purpose workhorses and offer a great mix of utility and sfaety for the money Volvo is long one of the leaders of safe cars, Subaru and AMC offered outstanding cars that give the security of AWD/4WD and fuel economy above thier competitors.
So depending on what you like there are many possible answers to your question.

THE best cars ever are Lexus rear wheel drive vehicles. Not Bugatti, not Ferrari; it is a Lexus.
Second, any small Toyota or Honda that is inexpensive. If you have a hundred million dollars it isn't difficult to make an exotic car. If the goal is to design a a beautiful, inexpensive, reliable car, that takes real talent.
