There are some rooms on IRC that have over 1000 people in them at once. They're typically file sharing rooms. Here's a list of the top 100 IRC channels based on current user count:

The top one right now has over 2000 people in the room. You need an IRC client like mIRC ( http://www.mirc.co.uk ) in order to chat on IRC.
As for the previous poster, I'm not aware of any single room on AOL that is even capable of holding 1000 people in it at once. The same goes for Yahoo. Yahoo Chat stinks anymore anyway with all the bots on there.
