It would have to be the St Ledger, the horse race for 3 year old colts and fillies and run at Doncaster every September. The race was first run in 1776.
It is not the oldest horse race though, that is the Newmarket Town Plate, the first race in the world to be ruin under written rules. It is run every October in Newmarket (though was run in August in 2006) and was first run in about 1665.
