If you are a "Trekkie " fan, then you also know that this phrase's origin is - that it is an abbreviated version of a traditional Jewish religious blessing. The phrase, accompanied by a hand gesture, came to a wider public in the Star Trek TV series as the greeting of the Vulcan people. There are claims about it not really being on the script but was adlibbed by Mr Nimoy during one of the episodes.
As for the proper response in the actual blessing, Repeat the same phrase back.

Some one should tell them that Live Long and Prosper is not a Star Trek original. In fact it was not even part of the Star Trek script, rather it was an adlib by Leonard Nimoy [Mr. Spock]. Nimoy has been a devout Jew his entire life and Live Long and Prosper is a bennediction that Rabbi's used to give over their congregation. In addition, the accompanying hand jesture is representative of the Hebrew letter Shin, which begins the word Shadai, one of God's sacred names. Just thought you might want to know the true origin of the phrase.

I am a Charter Trekker, having seen the originally broadcast episodes of TOS on TV in the '60's. I don't think there is a formal reply to "Live Long and Prosper", it is a greeting and farewell, just as many others, like Good 'Bye, So Long, Via con Dios, Shalom, Salaam, etc. It even has early variants itself, most notably, "Long Life and Prosperity!"
