Your microscope should have a scale in the ocular lens usually in micro-meters. Use this to get the size of your object on the slide. From what I know a pyrometer is used to measure temp. not the size of an object.

Use a hemocytometer. It has four square grids that are of a certain size. It is used to do cell counts, but you can get the size of objects laid on it and under the microscope. Any cell/bio lab has one.

Put an object of referance on the microscope, and it really depends on the power the microscope is on

What magnification?
Put an object of known size in the image?

I suppose you could calculate the size from the magnification factor, but you still need a reference.

i would assume you just put a ruler under the microscope....
