Many of these answers are good but another alternative depends on your router. If you have a modern router, ie Linksys, Belkin or Netgear rather than the standard ISP set up modem then you also have a built in firewall. In this router you can add any URL's to a block list and your router will filter these out so that they cannot annoy you any longer.

You need to run an anti spyware in Safe Mode.
You may need to switch off System Restore as back up files will be hiding in there, and it will just restore itself. Go to Start - programs - accessories - system tools - system restore.
Run your anti virus and anti spyware in Safe Mode. To get into Safe Mode repeatedly press the F8 key after the bios screen loads but before Windows loads. You'll know if you made it because the screen will be at 800 x 600 and your pc will be slow.
Then beef up your security. Its easier to prevent malware getting on to your sytem in the first place rather than trying to remove it. All the following programs are free for personal use;
You need all three anti spyware programs;

go to windows downloads type search VM890830-Vi.22.its a win 32 scanner for Trojans. 3 different settings on it.when you download click save option not run are you will have to keep going through same process.tip do it in safe boot,turn off system restore they hide in there until you run a restore

Get rid of Norton...it is hopeless. It's intrusive and awkward to use. It seems to be very problematic when it comes already installed...I had it on my first PC. I changed it asap. I've just gone with Mc Afee or Bitdefender8(a freebee)...also AVG since then...everyone has their favorite...always backed up with SPYBOT.
